(172) Substituting PEG-Lipid with Amphiphilic Polycarbonates in mRNA-Loaded Lipid Nanoparticles

Introduction: As the most clinically advanced formulations for mRNA delivery, lipid nanoparticles (LNPs) have gained traction with its impressive success of COVID-19 vaccines [1]. PEGylated lipid, a crucial component of LNPs, extends their systemic circulation by facilitating the particle stability in blood and avoiding the sequestration by mononuclear phagocyte system [2,3]. The polycarbonate-lipid hybrid mRNA nanoparticles (PC-HNPs) in this study were created by replacing ALC-0159, a PEG-lipid utilized in Pfizer-BioNTech's BNT162b2 vaccine with amphiphilic PEG-polycarbonate (PEG-PC) diblock copolymers.
