(185) An End-to-End High-Throughput Formulation Device Platform for mRNA & LNP Screening
Introduction: LNPs have been widely utilized in prophylactic vaccines and liver directed therapies; however, their applications in other areas remain limited due to challenges in payload design, LNP chemistry, composition, and formulation process optimization. To address these challenges and facilitate clinical translation, promising methodologies, such as barcoded in vivo screening and plate-based in vitro assays, have been adopted but often require labor-intensive, one-by-one formulation of LNPs. Therefore, high-throughput platforms are crucial to accelerating genetic medicine development
Learning Objectives:
Understand how different mixers can be optimized to produce LNPs with similar properties
Understand how different formulation platforms are translatable
A case study utilizing DOE to optimize LNP formulation
