(186) An alternative method to RNA lipid nanoparticle (LNP) production
Introduction: RNA-LNPs are a promising gene delivery modality for a wide variety of therapeutic applications. However, current methods of LNP production are limited by the need for complicated mixing and buffer exchange techniques (1). Here we present a method capable of producing RNA-LNP by loading pre-formed LNPs with RNA without the use of solvents or heating to encapsulate the RNA payload, giving essentially physically identical RNA-LNPs, with equivalent biological response, to those made by conventional LNP production methods.
Learning Objectives:
Understand the difference between alternative and conventional formulation methods.
Identify formulation parameters that are important to the alternative method.
Identify instances where the alternative method would be preferential to the conventional method.
