(201) Formulation strategies for stable lipid nanoparticles: insights and implications
Introduction: Lipid nanoparticles (LNPs) have gained widespread use following their approval as COVID-19 vaccines1. Despite their success, LNP stability remains a challenge, varying significantly based on their formulation2. This study aims to enhance mRNA LNP stability under various conditions, including elevated temperature, freeze-thaw cycles and lyophilization by systematically screening different formulation parameters such as pH, buffer systems, and excipients. The goal is to develop more stable LNP formulations that can improve the efficacy and shelf-life of future LNP vaccines and therapeutics.
