Immuno Delivery (Focus Group - ID)
Background and Purpose: A lipid nanoparticle-triple adjuvant complex (L-TriAdj) is being explored for intranasal pertussis vaccines to enhance mucosal immunity.[1,2] The lipid composition is critical to modulate cytotoxicity, uptake and cytokine expression from antigen presenting cells. The NPs were comprised of DDAB/DOPE/phospholipid/cholesterol, with NP size optimized by design-of-experiments methodology. The hypothesis was that phospholipid alkyl chain length (DPPC vs. DSPC), would be related to cellular uptake and stimulation of dendritic cell expression of immune stimulating cytokines.
