(373) Biodegradable silica-based ultra long-acting tirzepatide formulations for the treatment of obesity

Introduction: Over 30% of the global population are overweight or obese and alarmingly, projections suggest a rise to half of the world’s population by 2035(1). GLP-1RA are an effective solution for weight management, but rapid reversion of weight loss results in frequent and chronic therapies. Solutions are additionally challenging due to an ideally linear release profile, alleviating adverse effects. We have developed a silica composite based long-acting (LA) tirzepatide (TZP) depot formulation with stable release, promising better durability and compliance for patients, while minimizing treatment burden.

Describe the encapsulation of therapeutic agents through the use of silica nanoaggregates
Justify tunability of silica biodegradation rate to achieve long-term release of therapy
Understand how formulations can be screened rapidly, owing to simple accelerated dissolution methods
