(490) In situ forming implants: effect of drug-polymer-solvent interplay on PLGA degradation
Introduction: Little has been reported with respect to the interplay between the active pharmaceutical ingredient (API), the poly (lactic-co-glycolic acid) PLGA and the solvent on drug release and stability of in situ forming implants. The current research highlights the role of hydrogen bond forming groups on PLGA degradation. Leuprolide acetate, risperidone and paliperidone were selected as model drugs. The chemical structure of paliperidone is identical to that of risperidone except for the presence of a hydroxyl group. This makes for a unique comparison as risperidone has no hydrogen bond donors.
