Enhanced pDNA Delivery Mediated by Backbone-Degradable RAFT Copolymers

Introduction: Polymers present an attractive option for gene delivery due to their significant synthetic flexibility that allows their chemical composition to be precisely tailored. Condensing nucleic acids with cationic polymers forms a polyelectrolyte complex (polyplex), which can navigate across various cellular barriers to efficiently deliver the nucleic acid payload intracellularly. However, these highly charged polymers can also lead to cell cytotoxicity. As such, there is a strong unmet need to develop efficacious polymeric gene delivery vectors with lower toxicity and higher biocompatibility.
