mRNA-LNPs Microneedle Delivery in 3D Human Dermal Equivalent Model for Preliminary In-Vitro Testing

Introduction: Microneedles (MNs) are a cutting-edge intradermal delivery approach, enabling minimally invasive and efficient skin barrier penetration [1,2]. The proposed MNs encapsulate E-GFP mRNA lipid nanoparticles (LNPs) within the two main compartments of our PLGA microparticle (MP)-based MNs (the fast dissolvable tip and the slow-release MPs). In this study, we advanced MN characterization using an innovative skin-on-a-chip model based on an optically accessible platform integrating the human dermal equivalent (HDE) model[3,4].
