Dual Centrifugation: A Novel Perspective on LNP Formulation Development

Introduction: The administration of RNAs faces challenges such as a poor cellular uptake, rapid degradation and elimination, which limits their clinical application. Lipid nanoparticles (LNPs) proofed to be suitable drug delivery systems (DDS), to address these impediments. [1,2] Our work introduces dual centrifugation (DC) as a novel, rapid, and reproducible technique for formulating small, sterile, and highly concentrated LNPs. This method enables high-throughput screening with up to 40 vials per run and produces homogeneous RNA-loaded nanoparticles within just 7 minutes. [3]
