Multi-stage mixing creates high-efficiency core-then-shell DNA lipid nanoparticles

Introduction: DNA-loaded lipid nanoparticles (LNPs) offer unique advantages for treating chronic diseases, including months-long expression and cell-type-specific targeting through genetic promoters. However, their clinical translation has been limited by poor protein expression levels. Here, we introduce multi-stage mixing (MSM) microfluidics to create core-then-shell (CTS) structured DNA LNPs that dramatically improve transfection efficiency through controlled supramolecular assembly.

Describe how multi-stage mixing controls and create LNPs with unique core-then-shell structure
Compare core-then-shell versus traditional LNP formulation methods both in vitro and in vivo
Core-then-shell LNPs are able to load STING inhibitor and improve safety and efficacy.
