High-throughput screening of lipid nanoparticles for lung delivery of mRNA

Introduction: Lipid nanoparticles (LNPs) have paved the way for nucleic acid delivery. The fine-tuning of LNP composition enables targeted delivery to specific organs and administration via distinct routes1. Intranasal (IN) route provides access to the lungs without systemic exposure. However, determining an effective LNP composition to overcome barriers imposed by this route is a challenge. We utilized DNA barcodes for a high-throughput in vivo screening of a library of LNPs to lungs delivery following IN administration. The top-performing LNPs were further validated in terms of transfection efficiency.
