Elucidation of Tablet Dissolution and Functional Excipient Behavior with In Situ Exposure Imaging

Introduction: There is great interest in understanding the exact mechanisms of disintegration and dissolution of pharmaceutical dosage forms and in particular how the functional excipients behave to drive dissolution [1]. However given the range of timescales and the three-dimensional nature of the dissolution process, a mechanistic description of dissolution has proven to be extremely challenging to validate and verify. In this work we present dissolution and functional excipient characterization through development and implementation of time resolved 3D imaging of controlled release oral formulations.

To implement in situ µCT imaging for direct quantification of dissolution in ER and IR tablets.
Understand the role functional excipients play in controlling tablet dissolution.
Develop ultrafast in situ imaging that can be employed during traditional dissolution testing.
