A DNA-Guided Lipid Nanoparticle Platform for Targeted Drug Delivery Beyond the Liver

Introduction: To address the problem of hepatic accumulation upon systemic administration, there is a need for liver-detargeting formulations that offer tunability for active targeting. To meet this need, we present ‘LipiGo,’ a nanocarrier platform combining LNPs with DNA nanotechnology. By incorporating oligonucleotides into the LNP structure, LipiGo enables tunable size, charge, and biodistribution profiles. Additionally, through complementary DNA hybridization, it facilitates the precise attachment of targeting ligands, making it a versatile and powerful system for targeted delivery.
