The success of mRNA-based COVID-19 vaccines has highlighted the therapeutic potential of nucleic acid-based therapies. Lipid nanoparticles (LNPs) have evolved from research tools to clinically validated delivery systems, as seen with Onpattro® and mRNA vaccines. This study aimed to develop mRNA-loaded LNPs to modulate protein expression in skeletal muscle cells, serving as proof-of-concept for vaccine applications. Efficient transfection of muscle cells is crucial for local production of viral antigens and triggering a targeted immune response.
