Blood-brain barrier/glioblastoma interplay 3D in vitro model for (nano-)therapeutic screening

Introduction: Glioblastoma (GBM) remains an unmet clinical need. A key challenge is the limited ability of therapeutics to cross the blood-brain barrier (BBB) and reach the brain in effective doses. The in vitro selection of therapeutic candidates is often based on misleading and poorly predicted clinical responses, since current disease modeling toolboxes lack a close-to-native recapitulation of the blood-BBB-GBM bioarchitecture. We present a novel, reproducible 3D BBB/GBM model to stratify and screen BBB permeability and efficacy of (nano-)therapeutics, aiming to improve GBM treatment protocols.

Investigate how this model aligns with in vivo data and advances 3Rs-compliant drug testing.
