Engineering of mitochondria-enriched brain endothelial cell-derived extracellular vesicles

Introduction: We have previously demonstrated that microvesicles (MVs), a subset of large extracellular vesicles (EVs) contain mitochondria. MVs transfer their mitochondria into recipient brain endothelial cells (BEC) and increase recipient cell bioenergetics. Resveratrol (RES) increases mitochondrial biogenesis by activating peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α). We hypothesize that the quality and quantity of mitochondria packaged into MVs can be increased by activating PGC-1α in the donor BECs (Figure 1) and further increase the effects of mitochondrial transfer.
