Optimization of Nanoparticle Delivery of Nucleic Acids to Cells using Poly(glycerol esters)

Introduction: Poly(glycerol ester) (PGE) provides a next-generation polyplex for cell and gene therapy and ocular delivery. PGE nanoparticles have performance based on (1) the selection of the diacid structure and the amine functionality used; (2) the type and molar ratio of the polyol; and (3) the complexation buffer and parameters. A machine learning model of different polyplex preparation conditions was used to investigate the transfection performance of PGE nucleic acid delivery to HEK293T cells by predicting pareto-optimal polyplex preparation conditions to maximize transfection potential.
