Manufacturing and Process Scale-Up
The drug release kinetics of push-pull osmotic pump (PPOP) tablets are influenced by the hydration dynamics of both the push and pull (drug) layers [1–2]. Therefore, determining water uptake and its distribution within the PPOP system during dissolution is crucial for understanding the driving force that ensures linear drug release. This study examines the performance of glipizide PPOPs manufactured using different formulation strategies, with a particular focus on the impact of process parameters on water uptake, polymer layer swelling, and their effects on drug dissolution.
