(431) Cationic mucoadhesive Posaconazole NioTherms:A potential approach for management of Fungal Keratitis

Introduction: Fungal keratitis (FK) is a severe corneal infection, primarily affecting immunocompromised individuals causing vision loss. Posaconazole (PCZ), a broad-spectrum antifungal approved by the FDA, is used off-label for FK but faces delivery challenges due to poor corneal permeation and rapid ocular elimination. To address this, Niosome-like surfactant bilayer vesicles, NioTherms, were developed via a scalable, eco-friendly heat-mix method. These positively charged vesicles encapsulate PCZ within a stable bilayer, enhancing corneal permeability and retention when delivered as an ocular in situ gel.

Learning Objectives:

  • Conduct a scalable "heat-mixing" method for developing bilayered surfactant vesicles, NioTherms.
  • Demonstrate use of in silico tools for excipient selection and correlation with in vitro data.
  • Describe physicochemical parameters, pharmacokinetic and pharmacodynamic outcomes in small animals.
