(431) Cationic mucoadhesive Posaconazole NioTherms:A potential approach for management of Fungal Keratitis
Introduction: Fungal keratitis (FK) is a severe corneal infection, primarily affecting immunocompromised individuals causing vision loss. Posaconazole (PCZ), a broad-spectrum antifungal approved by the FDA, is used off-label for FK but faces delivery challenges due to poor corneal permeation and rapid ocular elimination. To address this, Niosome-like surfactant bilayer vesicles, NioTherms, were developed via a scalable, eco-friendly heat-mix method. These positively charged vesicles encapsulate PCZ within a stable bilayer, enhancing corneal permeability and retention when delivered as an ocular in situ gel.
Learning Objectives:
Conduct a scalable "heat-mixing" method for developing bilayered surfactant vesicles, NioTherms.
Demonstrate use of in silico tools for excipient selection and correlation with in vitro data.
Describe physicochemical parameters, pharmacokinetic and pharmacodynamic outcomes in small animals.
