(461) Identifying optimal process and formulation parameters for the TSWG of acetaminophen using L-HPC

Introduction:

Improving the flowability of poorly flowable active pharmaceutical ingredients is essential to ensure tablet weight and dose uniformity(1). In this work we aimed to identify the optimal process and formulation parameters for the application of L-HPC in twin-screw wet granulation of acetaminophen which results in improved powder flowability, granule and tablet properties.

Learning Objectives:

  • • Higher L/S ratios in TSWG of acetaminophen results in significant improvement in powder flow.
  • • Increasing LH-21 concentration results in a reduction of particle size and better tablet hardness.
  • • A 7.5% concentration of LH-21 and L/S ratio of 0.15 is recommended in the TSWG of acetaminophen
