(424) Breaking the Endosomal Barrier: Polymer-Enhanced Lipid Nanoparticles for Potent & Safe RNA Delivery
Introduction: Lipid nanoparticles (LNPs) are key non-viral carriers for nucleic acid therapeutics, but their efficacy is often limited by poor endosomal escape. While extensive efforts have focused on optimizing ionizable lipid structures, this critical barrier remains (1). We report the development of polymer-incorporated LNPs (p-LNPs), rationally designed to enhance endosomal escape and improve RNA delivery, addressing a major bottleneck in current LNP-based systems.
