High-resolution screening of barcoded lipid nanoparticles in tissue cleared mice

Introduction: Current LNP screenings lack multiplexing capabilities, only allowing for a small number of formulations to be tested in a single animal. This restriction increases costs, reduces throughput and requires the use of a large number of animals. Advances in molecular barcoding enabled the evaluation of multiple formulations in a single mouse(1). However, they still lack spatial information and rely on low-resolution imaging for selecting the region of interest(ROI). Advanced tools are needed to combine molecular and spatial information to provide detailed insights into biodistribution.

Conduct screening experiments with minimum number of animals.
